Description

Description: This assay employs a two-site sandwich ELISA to quantitate DSC1 in samples. An antibody specific for DSC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any DSC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DSC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DSC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Desmocollin-1 is a calcium-dependent glycoprotein that is a member of the desmocollin subfamily of the cadherin superfamily. These desmosomal family members, along with the desmogleins, are found primarily in epithelial cells where they constitute the adhesive proteins of the desmosome cell-cell junction and are required for cell adhesion and desmosome formation. The desmosomal family members are arranged in two clusters on chromosome 18, occupying less than 650 kb combined. Alternative splicing results in two transcript variants encoding distinct isoforms.Strongly expressed in epidermis, less in lymph node and tongue.The glycoproteins of 115 kD and 100 kD were named desmocollins to denote that they are involved in the adhesive function of the desmosomes
